ATM (ataxia telangiectasia mutated) and ATR (ATM and Rad3 related) are evolutionary conserved protein kinases that orchestrate cellular responses to DNA damage. One of their key substrates is the carboxyl tail of histone H2AX. Phosphorylated histone H2AX (aka γH2AX) tethers important DNA damage response (DDR) proteins to DNA double-strand breaks but its function during DNA replication is unclear. I will describe how a focused genetic screen with fission yeast has uncovered a critical requirement for formation of γH2AX during S-phase. |
