Elucidation of the activation mechanism of TGF-β-dependent gene expression

Transforming growth factor TGF-β is a multifunctional cytokine that is involved in the regulation of cell proliferation, immune functions, and malignant transformation of cancer. TGF-β binds to the cell surface receptors and regulates cellular signaling via intracellular transcription factor SMAD proteins and coactivator proteins such as CBP. In this study, we have successfully determined the structures of SMAD2-CBP complexes by x-ray crystallography.
TGF-β signaling is an attractive target for cancer therapy, because inhibitors of TGF-β may suppress the malignant transformation of cancer and activate immune functions. Therefore, elucidation of the specific binding mode between the transcriptional activator CBP and SMAD seems to be important for the development of anti-cancer drugs in the future.
